The Translational Genomics Core at Partners HealthCare Personalized Medicine supports expression-profiling analysis from total RNA. We offer expression analysis on Affymetrix and Illumina Microarray platforms. We are able to support samples many different species. Please contact Ashley Blau for chip availability.
We can accept either total RNA samples or samples already enriched for miRNA for miRNA analysis. All samples are tested for quality within 48 hours on Nanodrop and Agilent Bioanalyzer.
- Full-service option – Starting with total RNA, the full-service option includes labeling, hybridization, and scanning of the microarrays to produce a standard analysis report. Supported RNA labeling methods are NuGen, Ambion, Affymetrix IVT expression, and Genisphere.
- Hybridization and scanning service – If you have already prepared labeled RNA, we can hybridize it to the selected arrays and scan the arrays to produce a standard analysis report.
We also are able to provide NanoDrop and Agilent Bioanalyzer testing as a stand-alone service, should you wish only to have your RNA tested for quality.
RNA sample preparation
Be sure to take measures to ensure RNAse free conditions. Make your best efforts to preserve your RNA when isolating from tissues or cells. This may take the form of use of preservative, such as RNAlater or flash freezing in LN2 or, in the case of blood, use of PaxGene tubes. If using reagents such as TRIZOL, to extract your RNA, the prep must include a column purification (e.g., RNeasy) as a final step. Without this final column step, labeling reactions may fail or be attenuated, due to contaminants that may be present after organic reagent-based extractions.
Please treat samples using an in solution DNase treatment. We have found the on-column DNase digestion has not been suitable to eliminate all DNA.
